Abstract:Biorhythms, originating from the adaptive evolution of organisms to their natural environments, are ubiquitously observed across living systems on Earth. These rhythms play a crucial role in regulating various behavioral and physiological processes such as sleep, body temperature, blood pressure and hormone secretion.Rhythm disorders can negatively impact daily work, study and overall quality of life, and are related to an increase of the risk of various diseases.Conditions including hypertension, asthma, rheumatoid arthritis, and cancer, exhibit significant rhythmicity in their manifestations.Consequently, aligning drug administration strategies with these biological rhythms to maximize therapeutic efficacy has become a prominent focus in current research.Biorhythm-mimicking drug delivery precisely controls the release time and dosage of drugs to simulate the internal hormone secretion or gene expression rhythms of the body. For diseases with periodic fluctuations in symptoms or severity, such delivery systems can improve therapeutic outcomes and enhance compliance of patients. This review focuses on the recent advances in biorhythm-mimicking drug delivery and release systems and their applications in metabolic diseases, cancer, autoimmune diseases and neurological disorders. The potential challenges and future prospects are also discussed.
