Advances in cardiotoxicity assessment based on hSC-CMs under CiPA

Abstract:As drug cardiotoxicity studies have always attracted attention as an important part of drug safety evaluation,the US Food and Drug Administration(FDA)released a new evaluation system called comprehensive in vitro proarrhythmia assay(CiPA)in 2013.Its subgroup,the Stem Cell Working Group(SCWG)was established by the Health and Environmental Sciences Institute(HESI)to study the proarrhythmic effects of drugs using human stem cells-derived cardiomyocytes(hSC-CMs).Compared with in vitro models for predicting arrhythmogenic risk such as in vitro animal tissue models or other 2D cellular models,hSC-CMs have advantages in terms of species differences,patient specificity,and assessment of multiple ion channels.Based on this model,drug-promoted arrhythmogenic risk can be assessed using techniques such as MEA,VSO,and Ca2+transient monitoring.In this paper,we will highlight the research progress,limitations,and future perspectives of hSC-CMs in preclinical drug cardiotoxicity evaluation.
