Abstract:Stem cell-based regenerative medicine is one of the most compelling areas in the life sciences of the 21st century. However,in most cases, allogeneic transplantation can trigger an immune response, leading to an increase in allogeneic reactive memory T cells,antigen-specific antibodies, and reduced survival rates, thereby reducing treatment efficacy, inhibiting drug activity,or accelerating drug clearance, ultimately resulting in the loss of treatment effectiveness. This reaction targets allogeneic antigens which are recognized by the recipient's immune system as non self-antigens due to differences between individuals. Therefore, the failure of allogeneic transplantation caused by immunogenicity has become a key obstacle to the successful development of stem cell products. Therefore, understanding the causes and effects of immunogenicity is crucial for the application of stem cell therapy. The aim of this paper is to explain the mechanism of cell-mediated immune response and the common methods of immunogenicity detection in vitro,so as to provide a reference for the prediction of immune rejection of stem cells.
