Abstract:Schizophrenia is a class of severe mental disorder characterized by both positive and negative symptoms,as well as cognitive decline, which significantly impacts patients' health and interpersonal relationships. However, treatment outcomes are frequently compromised by poor medication adherence.Long-acting injectable antipsychotics(LAIs)have been shown to enhance adherence in patients with schizophrenia. On August 31,2021,paliperidone palmitate 6-month long-acting injectable(PP6M)was approved by the U.S.FDA for the treatment of adult schizophrenia.Clinical studies indicated that PP6M was as effective as paliperidone palmitate 3-month long-acting injectable(PP3M)in preventing relapse in patients with schizophrenia. This article provides a comprehensive discussion on the pharmacodynamics, pharmacokinetics, clinical efficacy and safety evaluation,cost-effectiveness,as well as drug interaction profile of PP6M.
