The mechanism study of Wumei Pills in inhibiting epidermal inflammation-mediated Th2-type inflammatory response for the treatment of atopic dermatitis

Abstract：Objective:To explore the mechanism of action of Wumei Pills in alleviating atopic dermatitis in mice.Methods:Forty BALB/c male mice were randomly divided into five groups:normal control group,model control group,positive control group(dexamethasone acetate),and high and low dose Wumei Pills groups.Except for the normal group,for which 20 μL of anhydrous ethanol solution was applied to the right ear of the mice,the mice in the rest groups were treated with 20 μL of 2 nmoL calcipotriol(MC903)solution repeatedly to the right ear for 10 consecutive days,while the left ear was untreated.The scratching behavior and the condition of ear skin damage of the mice were observed,the thickness difference and weight difference were measured between the left and right ear lobes of the mice,the histopathological changes in the epidermal layer of ear tissue was observed by HE staining,the number of mast cell infiltration in the dermis layer was observed by TB staining,and detection of TSLP,IgE,IL-4,IL-13,and IL-33 levels in mouse peripheral blood was performed by ELISA method.Results:In the Wumei Pills group,the AD-like symptoms of ear skin lesions in mice were significantly alleviated(P<0.05),scratching frequency was significantly reduced(P<0.001),the difference in thickness between the left and right ears was decreased(P<0.001),the weight difference was reduced(P<0.05),the ear skin lesion score was significantly lowered,the thickening of the epidermal layer was weakened(P<0.05),the number of mast cell infiltrations in the dermis was reduced(P<0.01),and the levels of peripheral blood factors such as IgE,TSLP,IL-4,IL-13,and IL-33 were decreased(P<0.05).Conclusion:Wumei pills can alleviate the symptoms of atopic dermatitis by inhibiting the release of TSLP/IL-33 factors from epidermal keratinocytes,reducing the production of Th2-type inflammatory factors and IgE,and further decreasing mast cell infiltration,thus exerting a therapeutic effect on atopic dermatitis.
