Research progress in new hypoglycemic agents as treatments for non-alcoholic fatty liver disease

Abstract:Non-alcoholic fatty liver disease(NAFLD)is the most prevalent chronic liver disease worldwide and is one of the common comorbidities of type 2 diabetes mellitus(T2DM).NAFLD exacerbates diabetes complications,while hyperglycemia accelerates the development of liver fibrosis,cirrhosis,and hepatocellular carcinoma.Insulin resistance(IR),which constitutes the first hit,runs through the entire course of NAFLD development.Given the key role of IR in NAFLD,many hypoglycemic drugs are being evaluated for their therapeutic effects on NAFLD.Recent research data indicate that new hypoglycemic drugs,while increasing insulin sensitivity,reducing liver enzymes,and decreasing body weight,also have a positive impact on liver lipid metabolism.This article reviews the effects of three major classes of new hypoglycemic drugs,namely incretin,sodium-glucose cotransporter 2 inhibitors,and peroxisome proliferator-activated receptor agonists,on the liver,in hopes of providing a scientific basis for their rational application in clinical practice and drug development.
