Trifluridine and tipiracil:a new drug for the treatment of gastrointestinal tumors

Abstract:Digestive system tumors are a serious threat to the health of Chinese people,among which gastric cancer,gastroesophageal junction cancer,and colorectal cancer have the highest incidence and mortality rates.Early diagnosis and early treatment have improved the treatment status of digestive system tumors in China,but the comprehensive treatment of patients with recurrence and metastasis is still a difficult point in treatment,especially the lack of safe and effective drugs for post-line treatment.Trifluridine and tipiracil(TAS-102)is a combination preparation,and in patients with metastatic colorectal cancer,the phase III RECOURSE and TERRA clinical studies showed that TAS-102 significantly prolonged the overall survival(OS)and progression-free survival(PFS)compared with the control group.For patients with metastatic gastric cancer and gastroesophageal junction cancer,the TAGS clinical study showed that the OS and PFS in the TAS-102 group were also significantly prolonged compared with the control group.The common adverse reactions of TAS-102 are mainly bone marrow suppression and gastrointestinal reactions,which are mostly grade 1~2 and can be tolerated.Domestic and foreign guidelines recommend that TAS-102 can be used as a posterior line option for metastatic colorectal cancer,gastric cancer,and gastroesophageal junction cancer.This article mainly introduces the mechanism of action,pharmacodynamics,pharmacokinetics,clinical trials and safety information of TAS-102,in order to facilitate its clinical application.
