Research of carcinogenic risk assessment for Pb and As in pheretima and scorpio based on in vitro PBET digestion/Caco-2 cell model

Abstract:Objective:To determine the contents of Pb and As in pheretima and scorpio by inductively coupled plasma mass spectrometry(ICP-MS)and establish in vitro physiologically-based extraction test(PBET)digestion/Caco-2 cell model to investigate the bioaccessibility of Pb and As.Methods:Carcinogenic risk(CR)method was used to assess the cancer risks owing to exposure to Pb and As.Results:After transportation by Caco-2 cell,the bioaccessibility values of Pb and As in pheretima were 6.21%~25.37%and 29.33%~65.36%,respectively,and were 26.89%~74.23%and 18.98%~56.78%in scorpio,respectively.The risk assessment results revealed that based on the total amount of heavy metals,the CR values of As in seven batches of pheretima were higher than 10-4,indicating a high cancer risk.After cell transportation,the CR values of Pb and As in all batches of pheretima and scorpio were below 10-4,indicating an acceptable cancer risk.Conclusion:This study establishes a novel model for risk assessment of heavy metals in pheretima and scorpio based on bioaccessibility,which provides a technical support for scientific and objective assessment of the health risks of heavy metals in traditional Chinese medicines and lays the foundation for the establishment of limit standard.
