Abstract:Objective:To compare the efficacy of inductively coupled plasma-mass spectrometry(ICP-MS)and atomic absorption spectroscopy(AAS)in determining the total arsenic content in clevidipine butyrate, and investigate the impact of different determination methods on the determination results of arsenic impurity content of the drug. Methods:Samples of clevidipine butyrate were fully digested using the microwave digestion technique. The arsenic impurities were then quantitatively analyzed using ICP-MS and AAS.The methods were evaluated based on their pre-treatment procedures, technical specifications, and methodological parameters, comparing their respective advantages, disadvantages, and applicability. Results:Both the microwave digestion method and the subsequent instrumental analyses proved to be efficient, requiring minimal time and reagents, and exhibited non-polluting characteristics, outperforming traditional methods and arsenic test.ICP-MS demonstrated superior linearity and sensitivity compared to AAS,particularly at low concentrations, where AAS showed reduced accuracy. The results of the sample determination using the two methods were essentially concordant. The results demonstrated that the ICP-MS method exhibits excellent detection performance and is particularly well-suited for the quantitative analysis of arsenic at trace to ultra-trace levels. Conclusion:The objective of this study is to assess and standardize the quality of clevidipine butyrate by evaluating the presence of arsenic impurities.ICP-MS is more suitable than AAS for the detection of arsenic impurities in drugs.
