Olanzapine/samidorphan:a new synthetic agent for the treatment of schizophrenia and bipolar I disorder

Abstract:Olanzapine is currently considered as one of the most effective medications for the treatment of schizophrenia.However,it has been found in research and clinical practice that many patients taking olanzapine gain significant weight and develop metabolic syndrome,which affects patient compliance.Samidorphan is a novel compound that,when combined with olanzapine,mitigates the risk of olanzapine-induced weight gain while maintaining its efficacy.Olanzapine/samidorphan was developed by Alkermes,an Irish biopharmaceutical company,and approved for marketing by the U.S.FDA on June 1,2021.This article provides an overview of the mechanism of action,pharmacokinetics,drug interactions,clinical efficacy,adverse effects,and precautions for use of olanzapine/samidorphan.
