Clinical research progress on Claudin 18.2 antibody-drug conjugates

Abstract：The tight junction protein subtype Claudin18.2(CLDN18.2)is exclusively expressed in normal gastric mucosal epithelial cells,where it plays a crucial role in regulating transmembrane transport of substances.However,CLDN18.2 is retained in the malignant transformation of normal tissues,and is widely expressed in gastric cancer,pancreatic cancer,breast cancer and other malignant tumors.Antibody-drug conjugates(ADC)targeting CLDN18.2 simultaneous exhibit high targeting selectivity of monoclonal antibodies and high anti-tumor activity of cytotoxic drugs,making them a promising new type of biologically targeted anticancer drug with great therapeutic potential.This article reviews the clinical trial progress of CLDN18.2 ADC,providing some references for the development of related drugs.
