Study on UPLC fingerprint and multi-component content determination of Kemin capsules

Abstract:Objective:To establish the UPLC fingerprint of Kemin capsules and screen its quality markers with chemical pattern recognition,so as to improve the quality standard of Kemin capsules.Methods:UPLC fingerprints were established for 25 batches of Kemin capsules,chemical pattern recognition[principal component analysis(PCA),hierarchical cluster analysis(HCA),orthogonal partial least squares-discriminant analysis(OPLS-DA)]was used to distinguish different origins and batches of Kemin capsules,and quality markers were screened.Results:The similarity of 25 batches of Kemin capsules was greater than 0.9,a total of 23 common peaks were calibrated,and 12 difference markers were screened by OPLS-DA method.Nine components including ephedrine hydrochloride,pseudoephedrine hydrochloride,methylephedrine hydrochloride,liquiritin,benzoylmesaconine,isoliquiritin apioside,benzoylaconine,benzoylhypacoitine,and ammonium glycyrrhizinate were identified as the quality markers of kemin capsules and the determination methods were established.The linear ranges were as follows:3.63~6.74,3.44~11.74,0.83~2.12,3.32~7.78,0.18~0.83,0.44~1.18,0.19~1.24,0.47~2.77,5.52~16.36 mg·g-1.Conclusion:The combination of fingerprinting,simultaneous determination of multi-component content and chemical pattern recognition can provide a more comprehensive evaluation of Kemin capsules and a theoretical basis for its pharmacodynamic material basic research and clinical application.
