Xuefu Zhuyu decoction decreases the risk of bleeding induced by different antithrombotic drugs

Abstract:Objective:To investigate the effect of Xuefu Zhuyu decoction(XZD)on bleeding induced by different antithrombotic drugs.Methods:The effect of XZD on the clotting time was observed by the capillary glass tube method in KM mice 1 h after a single administration.The effect of XZD on bleeding time in KM mice was determined by tail-tip amputation in KM mice 1 h after the final dose of 4 consecutive days.The combination of the traumatic blood stasis(TBS)model and warfarin was established in rats.After administration of XZD for 4 consecutive days,leg swelling was detected using the glass volume method,and ecchymosis was recorded 1 h after the final administration.The bleeding models induced by warfarin or low molecular weight heparin(LMWH)combined with aspirin were established in mice.After administration of XZD for 4 consecutive days,the bleeding time,activated partial thromboplastin time(APTT),and prothrombin time(PT)were measured 1 h after the final administration,and the international normalized ratio(INR)was also calculated.Results:Compared with the normal group,XZD prolonged clotting time 1.49-fold without affecting bleeding time.Concurrently,XZD reduced ecchymosis and leg swelling in rats treated with warfarin combined with TBS.XZD significantly decreased bleeding time by 47.07%,APTT by 35.54%,and PT by 29.53%in mice with warfarin-induced bleeding,while converting INR from a high bleeding risk range to a safe range.XZD significantly reduced bleeding time by 63.51%in mice with combination of LMWH and aspirin-induced bleeding and APTT unchanged.Conclusion:XZD reduced the risk of bleeding induced by different antithrombotic drugs.
