Progress in mRNA vaccine delivery systems for tumor gene therapy

Abstract:The utilization of mRNA cancer vaccines in tumor immunotherapy is becoming more prominent due to their notable attributes such as efficacy,safety,potential for further development,and cost-effectiveness.Nevertheless,challenges related to stability,innate immunogenicity,and in vivo delivery efficiency constrain the wide spread of mRNA cancer vaccines.The researchers enhanced the tolerance and stability of mRNA cancer vaccines by altering mRNA structure and optimizing delivery vectors.This paper provides a comprehensive review on the delivery systems of mRNA vaccines in tumor gene therapy,focusing on recent advancements and challenges associated with non-viral vectors like lipid nanoparticles,polymers,and cationic peptides.Additionally,the future potential of mRNA vaccines in cancer immunotherapy is also discussed.
