Immunological effects and preliminary mechanisms of Erxiangao formula on chemotherapy-induced leukopenia in mice

Abstract:Objective: To investigate the immunological effects of Erxiangao formula (EXG )on cyclophosphamide (CTX )-induced leukopenia in mice, and study its preliminary immunological mechanisms. Methods: After the model of leukopenia in mice was established by ip injection of CTX at the dosage of 200 mg· kg-1, the effect on leukopenia and immune function of EXG were evaluated by counting the numbers of peripheral white blood cells (WBC), bone marrow nucleated cells (BMNC)and T lymphocytes, measuring the capacity of splenocyte proliferation, calculating its thymus index and spleen index, determining the histology of femoral bone, and lastly measuring the level of cytokines by ELISA. Results: EXG could increase the numbers of WBC and BMNC in a dose-dependent manner, reverse the atrophy of thymus and spleen, and improve the femoral bone marrow morphology. Furthermore, EXG could also enhance the capacity of splenocyte proliferation and the total number of T lymphocytes and adjust the imbalance of T lymphocytes subtype ratio. Lastly, EXG could increase the levels of interleukin-2 (IL-2 )and interleukin-6 (IL-6 )in serum. Conclusion: EXG can promote the immune effects on chemotherapy-induced leukopenia in mice probably through increasing the level of cytokines, and these findings provided important information for the clinical use of EXG.
