Advance in the research of biological penetration enhancers in transdermal drug delivery system

Abstract:Transdermal drug delivery system (TDDS) has attracted much attention to pharmacy researchers because it has the characteristics of high local concentration,good controlled release effect and small side effects.It is an important clinical dosage form.The key to the success of TDDS is that drugs can transport through the skin effectively and reach the therapeutic target.Stratum corneum (SC) is the natural barrier of TDDS percutaneous absorption,so that most of the drugs are difficult to penetrate the skin to play a local or systemic treatment.Therefore,a variety of physical,chemical and pharmacy methods have been developed to increase the percutaneous absorption of drugs,in which the application of penetration enhancers is most extensive.However,the conventional chemical penetration enhancers (CPEs) are often limited by their inability to delivery large and hydrophilic molecules,and their incompatibility and local irritation issues.Therefore,there has been a search for biologicalpenetration enhancers (BPEs),which exhibit broad enhancing activity for more drugs without producing much irritation.In this paper,we present an overview of the types,characteristics,mechanism of action and the application of biological penetration enhancers for transdermal or dermal drug delivery systems,to provide the basis for selection of penetration enhancers and design of transdermal preparations.
