Effects of total flavonoids of Dracocephalummoldavica L· on learning and memory disorders of mice with dementia induced by D-galactose/sodium nitrite

Abstract:Objective:To observe the effects of total flavonoids of Dracocephalummoldavica L.on D-galactose/ sodium nitrite-induced learning and memory disorders in mice.Methods:Sodium nitrite and D-galactose were subcutaneously injected for 60 days to induce dementia model in mice.The effect of total flavonoids of Dracocephalummoldavica L (25,50 and 100 mg· kg-1) on the learning and memory disorders in mice were evaluated by Morris water maze,and the levels of superoxide dismutase (SOD),malondialdehyde (MDA),glutathione peroxidase (GSH-Px),interleukin-1β (IL-1β),tumor necrosis factor-or (TNF-ct) and nitric oxide synthase (iNOS) in the cortex were measured.Results:Compared with the model group,treatment with total flavonoids from Dracocephalummoldavica L.significantly improved the learning and memory deficits induced by D-galactose/sodium nitrite;the dose of 50 mg· kg-1 decreased the expression of MDA and IL-1β;the dose of 100 mg·kg-1 could obviously down-regulate the activity of SOD,and up-regulated the expressions of MDA,IL-1β and TNF-α.Conclusion:Total flavonoids from Dracocephalummoldavica L.can improve the ability of learning and memory in dementia mice and the mechanism is probably related to resisting oxidation damage and inflammatory reaction.
