Preparation of chlorophyll nonionic surfactant vesicles and characterization of their physical and chemical properties

Abstract:Objective:To prepare chlorophyll nonionic surfactant vesicles(Chl-NS) and study their physical and chemical properties.Methods:Chl-NS were prepared by thin-film ultrasonic dispersion method with Span-80,Tween-80 and cholesterol as membrane skeleton;the morphology of Chl-NS was observed by transmission electron microscope;the mean particle size,particle size distribution,Zeta potential and stability were measured by laser particle size analyzer;the vesicle encapsulation efficiency of chlorophyll and the changes of fluorescence intensity of vesicle-coated chlorophyll were determined by ultrafiltration-fluorescence spectrophotometry.Results:Chl-NS had spherical nuclear shell structure and were uniform in size with the mean particle size of (130.2 ± 10.0) nm,polydispersity index (PDI) of (0.240 ± 0.01),Zeta potential of(-30.0 ± 5.0) mV and encapsulation efficiency of (45.2 ± 2.1) %;the fluorescence intensity of the vesicles-coated chlorophyll aqueous suspensions was larger than that of the chlorophyll aqueous suspensions alone,with high stability.Conclusion:Chl-NS were prepared successfully with narrow size distribution,good stability,and strong fluorescence by thin-film ultrasonic dispersion method.The system is expected to be applied in the research of in vivo imaging system and has important clinical significance.
