Regulatory effect of fenofibrate on hepatic bile acids homeostasis and the possible mechanism

Abstract:Objective:To investigate the regulatory effect of fenofibrate on bile acids in mice fed with high fat diet.Methods:C57/BL mice were fed with high fat diet for 16 weeks,and then they were treated with or without fenofibrate (35 mg· kg-1,once daily) for 22 weeks.Blood glucose level,and the serum levels of total cholesterol,LDL cholesterol,ALT and AST were measured by commercially available kits.The livers were isolated for histological examination and gene expression analysis.Bile acids levels were detected by LC-MS technology.Results:Fenofibrate reduced the blood glucose,serum total cholesterol and LDL-cholesterol in mice with high fat diet.Meanwhile,it attenuated repression of the gene expression of Cyp7a1,FXR,Fgf15,JNK1,JNK2,and ERK1 in mice with high fat diet.LC-MS results showed that fenofibrate strongly increased DCA in the liver.Conclusion:Fenofibrate reduces cholesterol as well as blood glucose via regulation of bile acids pathway.Fenofibrate-induced abnormal accumulation of DCA may cause hepatic injury.
