Effect of cordycepin on apoptosis of human renal carcinoma cell line ACHN and its mechanism

Abstract：Objective:To study the effect of cordycepin on the proliferation and apoptosis of human renal carcinoma cell line ACHN,and explore its mechanism.Methods:CCK8 was used to detect the proliferation of ACHN cells treated with different concentrations of cordycepin for 24 h,and the inhibition rate and IC50 were calculated.Morphological changes were observed using an inverted microscope.Apoptosis of ACHN Cells which was treated with different concentrations of cordyceps for 24 h was detected by flow cytometry.BATMAN-TCM was used to preliminarily explore the possible mechanisms of cordycepin for inducing apoptosis.P-AKT and AKT were determined by Western blot.Results:Cordycepin resulted significant proliferation inhibition of ACHN cells.And it also effectively induced ACHN cells apoptosis with significant concentration dependence.The Western blot results showed that cordycepin dose-dependently inhibited activation of AKT protein in ACHN cells,which may be related to tits mechanism of apoptosis induction.Conclusion:Cordycepin can inhibit the proliferation of human renal cell carcinoma ACHN and induce its apoptosis,the possible mechanism of which may be inhibition of AKT phosphorylation,thereby inhibiting the activity of AKT pathway.
