Researches on obeticholic acid for the treatment of primary billiary cholangitis

Abstract:Primary biliary cholangitis (PBC) is a chronic and cholestatic liver disease,which is characterized by slow destruction of small intrahepatic bile ducts,stasis of toxic endogenous bile acids within the liver with progression to liver fibrosis and cirrhosis.Currently,PBC has an increasing prevalence worldwidely.Up to 40% of patients do not respond satisfactorily to the ursodeoxycholic acid (UDCA) therapy and these patients are at high risk for the disease progression.Obeticholic acid is effective in the treatment of patients with an inadequate response to UDCA or unable to tolerate UDCA,and obeticholic acid is well tolerated in these patients.In this review,epidemiology of PBC and the pharmacology,toxicology,efficacy and safety of obeticholic acid,latest approved by United States FDA,were summarized.
