Preparation and contents determination of the compound riboflavin in situ forming eye gel

Abstract:Objective:To explore the formulation and establish an HPLC method for the determination of the compound riboflavin in situ eye Gel.Methods:Using the gel state under 25 ℃ and 35 ℃ respectively,and the release test results in vitro as the indicators to select the appropriate formulation;Developed an HPLC method to determine the contents of riboflavin sodium phosphate and vitamin C in the gel.Results:The optimum formulation contained the same volume of methyl cellulose (0.1%),glycerin (VG∶ VW =1.4∶1) and sodium citrate (9%);Riboflavin sodium phosphate and vitamin C show good linearity at 0.04 ~ 0.20 mg· mL-1 and 0.02 ~ 0.06 mg· mL-1,and the average recoveries were 100.34% and 99.06%,respectively.The RSDs of precision were 0.055% and 0.98%.Conclusion:The presented formulation is appropriate and the assaying method is stable and reliable.
