Study on the synthesis of anti-hepatitis drug sofosbuvir

Abstract：Objective:To synthesis sofosbuvir,an anti-hepatiitis C drug,and optimize the preparing process.Methods:Sofosbuvir was prepared through the reduction,esterification,nucleophilic substitution,deprotection and nucleophilic substitution reaction using compound 1 as the starting material.Results:The structure of the target product was characterized by 1H-NMR and MS.The total yield was 37.7％.Conclusion:This improved method has the advantages of simpler operation,milder conditions and higher yield,which can be used for the industrial manufacturing.
