Establishment and biological characterization of sorafenib-resistant human hepatocellular carcinoma BEL-7402 cells

Abstract:Objective:The sorafenib-resistant human hepatocellular carcinoma BEL-7402 cell line was established and its main biological features were studied.Methods:CCK-8 assay was used to detect the inhibitory effect on cell proliferation.Coulter counting was used to assay proliferation rate.Cell cycle distribution and accumulation of intracellular reactive oxygen species (ROS) were analyzed using flow cytometry.Protein expression was detected by immunoblotting.Results:The sorafenib-resistant human hepatocellular carcinoma BEL-7402 cell line was established by concentration-stepwise treatment with sorafenib.The proliferation rate of the resistant cells slowed down.The cell cycle was changed,and the G1 phase cells increased significantly.In contrast to the sensitive cells,reductions of ROS levels and protein P53,and the decreased cleavage of the apoptotic marker protein PARP-1 were detected after the resistant cells were treated with the same concentrations of sorafenib.In addition,tetrandrine can reverse its resistance.Conclusion:The sorafenib-resistant human hepatocellular carcinoma BEL-7402 cell line was successfully established,which laid a good foundation for the research of the sorafenib resistance and related drugs.
