Effects of different blood sample treatment methods on biochemical indexes of rats

Abstract:Objective:To investigate the effects of different blood sample treatment methods on biochemical indexes of SD rats,and provide a reference for the application of plasma or diluted serum in pre-clinical safety test.Methods:Twenty rats were used for the study.Blood sample was collected via abdominal aorta,and then EDTA-K2 anticoagulated plasma,sodium citrate anticoagulated plasma,non-anticoagulant serum,and diluted serum (diluted with deionized water at volume ratio of 1 ∶ 1) were prepared.TBA-120FR was used for clinical chemistry analyses.Paired t-test was used for statistical analysis.Results:Compared with the serum test value,the AST,ALP,LDH and CK levels of the EDTA anticoagulated plasma were significantly reduced (P < 0.05).All the 13 indexes of the citrate anticoagulated plasma were significantly reduced,and the AST,TP,LDH and CK levels were still significantly reduced than the corresponding serum values even after being multiplied by the coefficient of 1.1.Compared with the serum value,the Alb level of diluted serum was increased and the TBil level was reduced (P <0.05).Conclusion:In pre-clinical studies,serum is recommended for biochemical determination.When plasma has to be used,sodium citrate anticoagulated plasma is not recommended,but EDTA-K2 anticoagulated plasma can be used.However,it should be noted that the AST,ALP,LDH and CK levels of plasma lack of comparability with the serum values.
