Tenofovir alafenamide, a nucleotide reverse transcriptase inhibitor
Abstract:Tenofovir alafenamide (TAF),a new kind of nucleoside reverse transcriptase inhibitor (NRTI),is the prodrug of tenofovir (TFV).TAF is a modified version of the drug Viread(R) (tenofovir disoproxil fumarate,TDF).Currently TAF has been approved by the Food and Drug Administration for treatment of HIV infection,and has potential of pre-exposure prophylaxis (PREP).This drug has also been recognized as a more effective anti-HBV drug globally.Compared to TDF,the clinical dose of TAF is lower while TAF has an easier entry to cells,which leads to greater antiviral efficacy and fewer side effects.The pharmacological actions,pharmacokinetics and clinical evaluation of tenofovir alafenamide were reviewed in this article.
