Effects of salvianolic-acid B on proliferation and extracellular matrix synthesis in rat adhesive fibroblasts induced by transforming growth factor-beta1

Abstract:Objective:To study the effects of salvianolic-acid B (SAB) on proliferation and extracellular matrix (ECM) synthesis in rat adhesive tissue fibroblasts (ATF) induced by transforming growth factor beta-1 (TGF-β1).Methods:The proliferation of ATF was detected by Cell Count Kit (CCK-8).The gene expressions of collagen I (COLI),fibronectin (FN),matrix metalloproteinase-9 (MMP-9) and tissue inhibitor of metalloproteinase1 (TIMP-1) were detected by real-time PCR.ELISA was employed to determine the protein levels of COLI,FN,MMP-9 and TIMP-1.Results:TGF-β1 of 10 ng· mL-1 could effectively up-regulate the COLI and FN expressions.SAB of 10 μmol· L-1 could suppress the ATF proliferation (P < 0.05) and down-regulate the expressions of COLI and FN in TGF-β1-stimulated ATF.Besides,SAB could also significantly up-regulate MMP-9 while down-regulate TIMP-1 in TGF-β1-stimulated ATF.Conclusion:SAB can inhibit TGF-β1-induced ATF proliferation.It can also restore the balance of MMP-9 and TIMP-1 expression in ATF,thus improve the ECM degradation,which may be the preventive and prophylactic mechanisms of SAB on post-operative adhesion.
