Research progress of compounds inducing readthrough of nonsense mutation

Abstract：Nonsense mutation is one of the most frequent causes of genetic disorders and cancer.Certain compounds,including aminoglycosides,non-aminoglycosides and small molecular weight compounds,show the ability to induce readthrough of nonsense mutation,which allows translation of full-length functional proteins.Although chemical-induced nonsense mutation readthrough has great potential as a treatment strategy for genetic diseases and cancer caused by nonsense mutation,few compounds show significant clinical benefit.Therefore,there is urgent unmet need to screen novel compounds with better therapeutic properties for moving readthrough strategy forward to clinic.Herein,we reviewed the research progress of compounds inducing nonsense mutation readthrough to provide information for the development of new pharmaceuticals.
