Folate-decorated nanostructured lipid carriers for pDNA targeting ovarian cancer

Abstract:Objective:To prepare folate (FA)-decorated nanostructured lipid carriers (NLC) as gene delivery system and evaluate the pharmaceutical and biological characteristics of FA-NLC/pDNA.Methods:First,FA-PEG2000-DSPE was synthesized.Cationic NLC was prepared by melt emulsion technology,and NLC/pDNA was obtained via electrostatic effect.Then,NLC/pDNA was coated by FA-PEG2000-DSPE.The characteristics of NLC,including particle size,zeta potential,entrapment efficiency,in vitro release,cytotoxicity and cell transfection were investigated and compared with Lipofectamin(R) 3000.Results:The average particle size of FA-NLC/pDNA was (143.8 ±5.1) nm,size distribution (PDI) was (0.19 ±0.04),Zeta potential was (12.1 ±2.9) mV,and the pDNA binding efficiency was (90.6 ± 2.7) %.The 60 h cumulative release of pDNA from FA-NLC/pDNA was about 80%,lower than NLC/pNDA and Lipofectamin(R) 3000.The cell viability assay showed that FA-NLC/pDNA exhibited low cytotoxicity to SKOV3 cells.After 48 h incubation,the gene transfection efficiency of FA-NLC/pDNA was significantly higher than that of NLC/pDNA and Lipofectamin(R) 3000.Conclusion:FA-NLC might be a promising non-viral gene delivery system with high efficiency and low cytotoxicity.
