Caspase-independent cell death induced by staurosporine in mouse T lymphocytes

Abstract:Objective:To investigate the caspase-independent death induced by staurosporine(STS)in mouse T lymphocytes,and analyze the death mechanism.Methods:The apoptotic rate and the changes in mitochondrial membrane potential of mouse T lymphocytes after treatment with STS(100 nmol·L~(-1))combined with or without z-VAD-fmk(20 μmol·L~(-1))were measured by flow cytometry and DNA ladder assay.The changes in apoptosis-related proteins such as AIF,Bax and cytochrome C were detected by Western blot analysis.Results:STS induced caspase-independent death of mouse T lymphocytes without showing a typical DNA ladder.The mitochondrial membrane potential was decreased,and the translocation of AIF from mitochondria to cytoplasm was earlier than that of cytochrome C.Moreover,Bax binding to mitochondria was necessary for the release of AIF and cytochrome C from mitochondria.Conclusion:The caspase-independent apoptosis may be an alternative pathway of caspasedependent apoptosis in the mouse T lymphocyte death induced by STS,which may play a critical role for the maintenance of immune cell homeostasis.
