Progress in application of dendrimer polyamidoamine as drug carrier

Abstract:Dendrimer polyamidoamine(PAMAM) is a hyperbranched and three dimensional macromolecule which possesses properties of monodispersion, non-immunogenicity, low cytotoxicity and biodegradation. Recently, as drug carrier, it has been paid more attention in pharmaceutical science. In this review, we described drug loading mechanisms as well as the applications of dendrimers in different drug administration routes such as vein injection, oral administration, transdermal delivery, ocular delivery and so on. It has been found that the drug loading mechanisms are physical encapsulation and electrostatic interaction, covalent combination. PAMAM as drug carrier could increase the solubility of the drug, delay drug release, increase the skin permeability coefficient, prolong the residence time in cornea and increase the bioavailability of the drug. It might be a potential and novel drug carrier.
