Biological degradation of an absorbable polysaccharide agent

Abstract:Objective:To study the degradation of starch-derived absorbable polysaccharide hemostat (SAPH) in organism and tissue,and evaluate its safety in surgery. Methods:SAPH product Perclot~(TM)(2μg) was injected into brain or abdominal cavity. After 30,60 or 120 minutes,the supernatants of brain homogenate and ab-dominal cavity lavage fluid were extracted. The Perclot~(TM)concentration of samples was detected by iodine solution reaction with standard curve. In vitro,2μg Perclot~(TM)was mixed with the homogenate supernatants of 1 g brain, liver, kidney or lung tissues,respectively. Additionally,0.01~1μg Perclot~(TM)was mixed with the homogenate superna-tants of 1g brain tissue. After being incubated at 37℃ for 30 minutes, the remnant starch concentration was detec-ted. Results:After being injected Perclot~(TM)for 30, 60 or 120 minutes, their remnant ratio were 38.62% ,6.48% and 0.85% in brain,and 64.08% ,33.36% and 1.78% in abdominal cavity, in vitro the Perclot~(TM)was degraded more than 50% by incubating with the tissues homogenate for 30 minutes. Liver tissue was prior to the others in starch degradation. Within 30 minutes, 50% of Perclot~(TM)above 0.1μg·mL~(-1)was degraded in brain tissue homoge-nate. Conclusion:The SAPH could be degraded promptly in brain or abdominal cavity of rats. In vitro, the degradation was found in tissues homogenate such as brain, liver, lung and kidney. SAPH can be safely applied in surgery.
