Preparation of glipizide extended-release dropping pills and its in vitro release behavior

Abstract:Objective:To prepare the glipizide extended-release dropping pills (Gli SDP) and investigate the in vitro release behavior. Methods:Gli SDP were prepared by solid dispersion technique using polyethylene gly-col (PEG) and stearyl alcohol as carrier materials,and the release profile of glipizide from Gli SDP was assessed by linear and nonlinear least square regression methods respectively. Results:According to the r~2(determination coeffi-cient) and AIC(Akaike's informulation criterion),the zero-order and Higuchi models were the optimal fitting mod-els. The cumulative release rate of glipizide from GIi SDP in 16 hours was over 95%. Conclusions:The release of glipizide from Gli SDP in vitro was nearly at constant rate, and more complete than that from commercial prepara-tions,and diffusion was the main release mechanism.
