Determination of lysophosphatidyl cholin in soybean phospholipid by HPLC-ELSD assay

Abstract:Objective: To determine the concentrations of lysophosphatidyl cholin (LPC) in soybean phos-pholipid by HPLC-ELSD method. Methods: The separation was performed on a NH_2 column ( 150 mm×4.6 mm,5 μm) with the elution mobile phase consisting of solvent A (methanol-chloroform, 97:3 ) and solvent B (2% oxalic acid-ethanol, 16: 84). The flow rate was 0.8 mL ·min~(-1) and the detector was evaporative light-scattering detector (ELSD). The column temperature was set at 35 ℃. The drift tube temperature of ELSD was 80 ℃ and gas flow rate of the ELSD was 2.0 L·min~(-1). Results: The linear range was 0.2~3.2 mg (r=0.999 3) with the average re-covery rate was 100.1% (n =9). The detection limit was 80 ng for LPC. Conclusion:The method is rapid,accu-rate and reliable, and can be used to determine the lysophosphatidyl eholin in soybean phospholipid.
