Abstract：Objective: To establish a HPLC method for the determination of irbesartan and hydrochlorothia-zide tablets and the related substances. Methods:A gradient HPLC method was used for the determination of the irbesartan and hydrochlorothiazide tablets and its related substances. Its major related substances were also subjec-ted to on-line LC-MS/MS and/or post preparation off-line ~1H-NMR studies. Results:Irbesartan,hydrochlorothiazide and their related substances were separated by the established HPLC and LC-MS/MS conditions. The blank sam-ples and solvents did not interfere with the assay of the related substances. The major related substances of irbesar-tan and hydrochlorothiazide tablets were characterized by LC-MS/MS and ~1H-NMR. Conclusion: The gradient HPLC method was specific,accurate and suitable for the measurement of irbesartan and hydrochlorothiazide tablets and its related substances.
