Study of lignans of Rhizoma Typhonii on the expression of TRAIL and its receptors in gastric carcinoma cell line (SGC-7901)

Abstract:Objective: To study the effect of lignans of Rhizoma Typhonii on the expression of TRAIL and its receptors in gastric carcinoma cell line (SGC-7901). Methods: The proliferation of SGC-7901 cells was meas-ured by MTT assay after exposure to extracts of Rhizoma Typhonii at concentration 0.026~26 μg·L~(-1) for 24 h. The morphology of cell apoptosis was observed by Hoechst33342 fluorescent staining. TRAIL and TRAIL-Rs mRNA expression in SGC-7901 cells were determined by RT-PCR. Results: The inhibition rate was 7.35%, 16.11%, 27.18% and 50.58% at the different concentration 0.026~26 μg·L~(-1) extracts of Rhizoma Typhonii, respectively. Interestingly, the expression of TRAIL,TRAIL-R1 and TRAIL-R2 was significantly higher in the experiment groups at concentrations than control group. Conclusions: The up-regulation of TRAIL/TRAIL-R1 and TRAIL-R2 by Lig-nans of Rhizoma Typhonii could be involved in the induction of apoptosis in SGC-7901 cells.
