Effects of catechin on functions of fibroblast-like synoviocytes under the stimulation of IL-1α in adjuvant arthritis rats

Abstract:Objective: To study effects of catechin (Cat) on fibroblast-like synoviocytes (FLS) under the stimulation of recombinant rat interleukin-l-alpha (rIL-1α) and investigate the mechanism. Methods: Freund's adjuvant (FCA) was used to induce adjuvant arthritis (AA) in rat. The FLS from AA rats was collected and incu-bated. Synoviocyte proliferation was determined by 3-(4, 5-dimethyhhiazal-2yl) 2, 5-diphenyhetrazoliumbromide (MTT) assay. The levels of PGE_2 and cAMP were determined by radioimmunoassay (RIA). Results: IL-1α at the optimal concentration of 10 ng·mL~(-1) significantly reduced the level of cAMP and increased the level of PGE_2. Cat (100, 50, 25 mg·mL~(-1)) inhibited the proliferation of FLS and decreased the level of PGE_2 and increased the lev-el of cAMP. Conclusion: Cat showed a capacity to reverse the rIL-1α stimulation on the FLS.
