In vivo distribution of gatifloxacin polybutylcyanoacrylate nanoparticles in mice

Abstract:Objective: To investigate in vivo distribution of gatifloxacin polybutylcyanoacrylate nanoparticles (GPN)in mice. Methods: Male mice were separated randomly into three groups and received single intravenous injection of gatinoxacin physiological saline solution(I),GPN without modification(II)and GPN modified with polysorbat-80(T-GPN,Ⅲ)via the tail vein respectively. The concentration of gatifloxacin in plasma, liver, kidney, spleen and lung were determined by HPLC and its distribution characteristics were investigated. Results: The peak concentrations of gatifloxacin in the lung of group Ⅰ,Ⅱand Ⅲwere 17.33,10.13 and 26.99 μg·g_(-1).The area under the concentration-time curve between 0 and 48h(AUC_(0-t))were 85.99,100.00 and 242.34 mg·kg_(-1)·h. Conclusion: The GPN modified by polysorbat-80 showed the characteristics of lung targeting.
