Abstract:Ceftriaxone sodium and sulbactam sodium for injection [2:1, CTZ/SBT (2: 1)]is a new antibiotic compound for drug-resistant bacteria. In in vitro pharmacological studies, MIC_(50) and MIC_(90) of the combination for beta-lactamase-producing bacteria are significantly lower than those of CTZ alone. This antimicrobial advantage has also been proved in infected rats. The safety of the compound has been demonstrated by toxicity assays in rats and dogs after systemic administration for 30 days. Tolerance experiment shows its safety ranges about 5.0 g in health volunteers. In phase I trials, no statistical differences have been found in the pharmacokinetic parameters of CTZ and SBT between administrations alone or in combination. The multi-center, randomized, controlled, blinded clinical trials have displayed the potency of the compound. In acute bacterial respiratory and urinary infections, the efficiencies were 88.33%and 90.00%,the clearance rates were 100.00% and 91.67%,and the sensitive rates were 100.00% in both infections. Respectively:the overall rate of adverse effects was 5.79%.In conclusion, the compound is a safe and effective treatment for bacterial infections, especially for the treatment of beta-lactamase- producing bacterial infections in clinic.
