GFP-1, exo-polysaccharide from Grifola frondosa, inhibits a-glucosidase

Abstract:Objective:To investigate the inhibition of a-glucosidase by GFP-1, exo-polysaccharide from Grifola frondosa. Methods:Exo-polysaccharide was prepared by submerged mycelia-um culture of Grifola frondosa, and GLP was isolated from the exo-polysaccharide via deproteinization, decoloration and dialyse. The inhibition of α-glucosidase was determined under standard system. Results: GLP-1 inhibited a-glucosidase with the IC_(50) value of 35. 79 mg·mL~(-1) under standard system. The action reached the maximum with 4min. It had thermal and pH stabili-ty; the Michaelis constant (Ki) was 3.4 × 10~(-4) mol×L~(-1) Lineweaver-burk plot showed that the inhibition of α-glu-cosidase activity was non-competitive. Conclusion:GLP-1 has a high affinity and inhibiting activity for α-glucosi-dase. It may inhibit sugar absorption.
