Protective effects of Xiaoketongluo granula on diabetic peripheral neuropathy in rats

Abstract:Objective:To observe the effects of Xiaoketongluo granula (XKTL) on the diabetic peripheral neuropathy ( DPN) in rats,and to investigate the possible mechanism. Methods:Diabetes was induced by streptozo-cin(STZ). Diabetic rats were treated with oral XKTL at 3 doses. After 8-week XKTL treatment,the effects on the FBG,HbA1c,aldehyde reductase ( AR) ,Na~+-K~+-ATPase and morphological changes were observed. The conduc-tion velocity of sciatic nerve and the pain threshold detected by illumination test were observed,respectively,at the end of the 4th and 8th week. Results: As compared with diabetic control,XKTL reduced AR activity and increased Na~+-K~+ -ATPase activity in sciatic nerves of diabetic rats. Meanwhile,it obviously improved the nerve conduction velocity and the pain threshold,and alleviated the pathological changes ( myelin sheathe swollen of the sciatic nerves) in diabetic rats. Conclusion: XKTL alleviates the lesion of DPN in rats; this may relate to the improvement of metabolic disturbance and the inhibition of the polyol pathway.
