Pemirolast potassium, an inhibitor of type Ⅰ allergy

Abstract:Pemirolast potassium is an inhibitor of type Ⅰ allergy. Preclinical studies indicate that it can sta-bilize the cell membrane of mast cells, inhibit the inflow and release of Ca~(2+), inhibit the activation of phosphodies-terase and increase the intracellular cAMP levels. It also dose-dependently suppresses the release of allergic media-tors induced by the antigen-antibody reaction, such as histamine, leukotrienes, prostaglandin D_3, thromboxane B_2 and so on. Clinical studies suggest that pemirolast potassium can prevent and cure variation asthma, anaphylactic rhinitis, anaphylactic conjunctivitis and atopic dermatitis with little untoward reactions. This article provides an overview of the physicochemical properties, action mechanisms, preclinical study, pharmacokinetics and pharmacody-namics, clinical study and the untoward reactions of pemirolast potassium.
