New synthesis process of salmeterol xinafoate

Abstract:Objective: To study a new synthesis process for preparing salmeterol xinafoate. Methods: Started from salicylaldehyde and 4-phenyl-1-butanol, the target compound was synthesized from salicylaldehyde via several steps involving acetylization, condensation, reduction, and catalytic hydrogenolysis. Results: The total yield was 31.5%. Structures of the target product and key intermediates were verified by IR and ~1H-NMR. Conclusion: This optimal synthetic procedure has the advantages of lower cost and higher yield, and is worthy to further industrialized manufacture.
