Protection of scutellarein on cerebral ischemia in rats

Abstract:Objective: To investigate the protective effect of scutellarein on rat cerebral ischemia. Methods: After oral administration of scutellarein 100, 50 or 25 mg·kg~(-1) for 7 days, focal cerebral ischemia was induced in male Wistar rats by middle cerebral artery occlusion with the method of electric coagulation, and 24 hours later, the neurological deficit score and the cerebral infarct volume were assessed. Besides, in rats undergone same medication schema, global cerebral ischemia was induced by ligation of bilateral carotid and the pathological changes were assessed meanwhile the water content and Na~+ , K~+ concentration in brain were also measured. In both experiments, 100 mg·kg~(-1) scutellarin was tested as positive control. Results: Scutellarein ameliorated the neurological behavior and restricted the infarction development in MCAO rats in a dose-dependent manner. Also, it reversed the brain edema, the ionic disorder and the pathological changes in global cerebral ischemia. Conclusion: Scutellarein has protective effect, better than scutellarin, in rat cerebral ischemia.
