Identification of clubroot resistance in radish germplasm resources and QTL-seq mapping

Abstract:Clubroot disease,caused by Plasmodiophora brassicae Woronin,is a globally distributed soil-borne disease that seriously threatens the sustainable development of the Brassica vegetable production.To clarify the resistance levels of radish germplasm resources and identify resistance loci,this study systematically evaluated the clubroot resistance of 40 radish varieties through integrated field natural infection and seedling-stage artificial inoculation methods.Segregating populations constructed from resistant and susceptible parents were subjected to QTL-seq-based mapping analysis.The re-sults demonstrated that one local variety and four hybrids exhibited immunity(Ⅰ),one local variety and three hybrids showed high resistance(HR),and three local varieties showed disease resistance(R).QTL-seq analysis identified four re-sistance-associated QTLs on chromosomes 1,2,and 5,with the major-effect locus rs2.1 located on chromosome 2.This research successfully screens multiple elite resistance sources and maps clubroot resistance loci in radish,providing cru-cial scientific foundations for disease-resistant breeding and gene cloning.
