KASP marker for identification of seed purity in watermelon special bottle gourd rootstock F1 hybrid Suzhen No.1

Abstract：In order to identify the seed purity of the hybrid seeds of Suzhen No.1,a special watermelon bottle gourd type rootstock accurately and efficiently,two stables polymorphic KASP markers(SNP1101 and SNP1102)were screened and obtained by using Suzhen No.1 and its parental inbred lines as experimental materials,combined with simplified resequencing and KASP technology. These two KASP markers were used to detect the genotype of 537 individual plants belonging to Suzhen No.1 and the accuracy of these markers were rigorously validated by combination with the field phenotypes. The results showed that the seed purity of this batch of Suzhen No.1 was 99.44％,and the identification results of the two pairs of KASP markers were consistent with the result of field phenotype identification,which could be used to distinguish Suzhen No.1 from other bottle gourd-type rootstock varieties. Above results can provide a convenient method for efficiently and accurately identifying the bottle gourd-type rootstock varieties and seed purity.
