In vitro culture of small fruit type watermelon cotyledon node

Abstract:In order to explore the small fruit watermelon tissue culture techniques and establish the aseptic system process,the small watermelon inbred line K1 was used as experimental material,the small watermelon seed germination conditions in vitro and effects of plant growth regulators on adventitious bud regeneration from cotyledonary node were investigated in this study.The results showed that the best soaking and sterilizing methods were that the seeds were sterilized in 3% NaClO for 20 min,then soaked in sterile water for 6 h,after removing the seed coat,the seeds were sterilized in 70% alcohol for 60 s,then soaked in 3% NaClO for 20 min,finally the seeds were washed in sterile water for 3 times.The highest seed germination rate appeared in the 1/8 MS media.The optimal medium for watermelon cotyledon adventitious shoot induction was MS medium supplemented with BA 2 mg· L-1,NAA 0.1 mg· L-1.
