A study of factors affecting anther culture of watermelon

Abstract：The purpose of this experiment was to explore the bud morphological index,high temperature pretreatment and medium hormone formula of watermelon anther culture.The growth of callus was observed and analyzed through bud grading,high temperature pretreatment and different induction medium.The results showed that when the flower buds and anthers were in medium size,it was the suitable period for anther inoculation of the experimental materials.The anther of watermelon was treated of 3 days at 33 ℃,and then 14 days at 25 ℃,then the anther callus could be stimulated.The induction rate of callus was higher when the hormone ratio of induction medium was 1.0 mg· L-16-BA +0.4 mg· L-1NAA and 1.5 mg· L-16-BA +0.2 mg·L-1 NAA.
