Ultramicroscopic observation of protein crystals in leaf bades of Solanum melongena

Abstract:Transmission electron microscopy was applied to characterize protein crystals in leaf blades of Solanum melongena,with the aim to understand whether the occurrence of protein crystals has tissue and intracellular compartment specificity and what shape the crystals take.It was demonstrated that protein crystals occurred in all types of cells.Protein crystals were present only in peroxisomes and occupied most of their volume,and no protein crystals were found in other intracellular compartments.Protein crystals had varied sizes,dependent upon the size of peroxisomes where they were located.The 2-D images of protein crystals were trapezoid,parallelogram,rectangle,square,pentagon,and parallelepiped.These 2-D images suggested a parallelepiped 3-D images of protein crystals.The sections of the crystals revealed a striped pattern.In conclusion,the occurrence of protein crystals in Solanum melongena leaf blades did not have tissue but intracellular compartment specificity,and the crystals took a parallelepiped shape and their sections had a striped ultrastructural pattern.
