DNA extraction from different types of watermelon samples

Abstract:The different types of watermelon samples of seed, cotyledon, hypocotyl, seedlings, fresh true leaf and dry true leaf were used as materials to compare the DNA extraction effect. The results showed that DNA extracted from fresh leaves and dry leaves was the highest concentration with 1 039.7, 1 130.2 ng ·μL-1 respectively, it also showed that the extraction efficiency of DNA from fresh leaves and dry leaves was not quite different, correspondingly, DNA extracted from cotyledon, hypocotyl, and seedlings were moderate concentration with 434.3, 406.3, 429.0 ng ·μL-1, DNA extracted from seed was the lowest concentration with 55.1 ng ·μL-1. dCAPS amplification and enzyme digestion showed that DNA extracted from all watermelon samples could be amplified and be digested by incision enzyme successfully, and also showed DNA extracted from different samples could meet the requirements of dCAPS markers research.
